I have a 10 day old pug and when they were feeding tonight I noticed a lump on the side of the puppy's rectum.
I cleaned it with a little warm water and it burst, it looked like pus and a little blood came out. I put a little neosporin on it.
Should I worry about this, I really didn't want to take a 10 day old puppy out of the house yet!

It's difficult to say whether or not this needs veterinary attention, but if you are in any doubt then take your puppy to your vet.
Little ones like this can get very sick, very fast and it's always better to be safe than sorry.
As the lump burst and leaked the pus/blood then as long as you keep it clean and dry (and using a little Neosporin can't hurt I would think), hopefully it will heal up. However, the pus indicates infection was present, so if it starts to increase in size, looks red or has pus building up again, or seems to be painful you need to get your puppy examined by your vet as antibiotics are most likely needed.
I'm not a vet and I can really only recommend that you take your own vet's advice on this, so if you are at all concerned make an appointment for your puppy.
