It sounds as though your pup may have a swollen lymph node in his neck. This is usually caused by an infection of some kind and needs to be evaluated by a veterinarian. Antibiotics are the most likely treatment if it is infection.

There is also a condition known as 'Puppy Strangles' whose main symptom is swollen lymph nodes in the neck. It's an immune system condition and needs to be treated with antibiotics and corticosteroids to reduce the swelling.

If your puppy isn't eating or drinking he is at serious risk of dehydration. Given his symptoms he needs to be seen by a veterinarian immediately so that you can find out what exactly is causing the problem and get treatment started.
